摘要
目的以siRNA抑制结肠癌HT-29细胞内survivin的表达,探讨HT-29细胞对顺铂(cisplatin,cDDP)的增敏效果。方法转染siRNA于HT-29,检测survivin siRNA处理对survivin、Bax、Bcl-2蛋白表达及胱天蛋白酶-3(caspase-3)活性的影响。cDDP处理后,以四甲基偶氮唑蓝比色(methyl thiazolyl tetrazolium,MTT)及酶联免疫吸附剂测定(enzyme-linked immunosorbent assay,ELISA)法分析HT-29细胞增殖抑制及凋亡情况。结果 Survivin siRNA下调survivin、Bcl-2蛋白表达,上调caspase-3活性及Bax蛋白表达。Survivin siRNA还可增强cisplatin对HT-29的生长抑制效果及提高cisplatin所诱导的HT-29凋亡发生水平。结论 Survivin siRNA可提高HT-29对化疗药物cisplatin的敏感性。抑制survivin可能是一个重要的、可行的结肠癌治疗标的。
Objective To silence survivin gene expression with siRNA and investigate its effect on enhancing the chemosensitivity to cisplatin(cDDP) in HT-29 colon cancer cells. Methods SiRNA was transfected into HT-29.Effect of survivin siRNA treatment on survivin,Bax and Bcl-2 protein expressions and caspase-3 activity were assessed.After cisplatin treatment,methyl thiazolyl tetrazolium(MTT) assay and enzyme-linked immunosorbent assay(ELISA) were used to evaluate the proliferation inhibition and apoptosis of HT-29 cells. Results Survivin siRNA downregulated the survivin and Bcl-2 protein expressions,and upregulated caspase-3 activity and Bax protein expression.Survivin siRNA could enhance both the inhibitory effect of cisplatin on growth of HT-29 cells and the level of apoptosis induced by cisplatin in HT-29 cells. Conclusion Survivin siRNA can enhance the chemosensitivity of HT-29 cells to cisplatin.The inhibition of survivin may be an important and feasible therapeutic strategy for colon cancer.
