摘要
用PR-HPLC法测定丁公藤注射液有效成分总东莨菪内酯在小鼠血浆中的浓度。色谱条件:SpheriSorb_C18不锈钢柱,流动相为甲醇-水-冰醋酸(38∶62∶0.19),检测波长345nm。血浆样品经酸水解后用氯仿萃取,50℃水浴,氮气吹干,甲醇超声溶解后进样。本法具有快速、简便、灵敏、准确等特点,已经用于总东莨菪内酯血药浓度测定及药代动力学研究,结果满意。
The plasma concentration of total scopoletin in Dinggongteng Injection was determined by RP_HPLC. The chromatographic conditions were as follows: stainless column Spherisorb C 18 ; mobile phase:methanol_water_glacial acetic acid (38∶12∶ 0.19 ); detection wave length 345nm. The plasma sample was extracted by chloroform after acid hydrolysis, dried by nitrogen on a water bath at 50℃, and dissolved with methanol. This method is rapid simple, sensitive and accurate, and has been used in plasma concentration determination and pharmacokinetics of total scopoletin.
