摘要
[目的]为海藻糖的科学生产提供参考。[方法]对以红薯淀粉为原料发酵生产海藻糖的过程进行研究,通过正交试验确定海藻糖合成酶产生菌的最佳培养基配方,使用最佳培养基来确定最佳发酵时间、发酵温度、pH、装液量等最佳培养条件。[结果]以食尼古丁节杆菌为生产菌,得到海藻糖合成酶的产酶最佳培养基配方为:麦芽糖4%、蛋白胨0.5%、牛肉膏0.6%、K2HPO40.08%、NaH2PO40.12%。确定了以红薯淀粉为原料发酵生产海藻糖的培养条件为发酵时间48 h,最适发酵温度38℃,最适pH 7.0,最适装液量200ml/500 ml。[结论]确定了红薯淀粉发酵生产海藻糖的小试最佳培养基配方和发酵条件,为红薯淀粉发酵生产海藻糖后期的中试和工业化生产提供了科学依据。
[Objective] The reference for the scientific production of trehalose was provided through the research.[Method] The fermentation process of sweet potato starch as raw material for trehalose production was experimented and the component of medium that was used for the bacterial strain producing trehalose synthase was optimized based on the orthogonal test so that the best culture condition such as fermentation time and temperature,pH value and culturing volume in the optimal medium was determined [Result] The best medium component for Arthrobacter nicotinovorus as the strain producing trehalose synthase was: the amount of maltose was 4%;the peptone,0.5%;the beef extract,0.6%;K2HPO4,0.08%;NaH2PO4,0.12%.And also,the best culturing condition for the trehalose production from the sweet potato starch fermentation was the fermentation time was 48 hours;the fermentation temperature,38 ℃;the pH value,7.0;the liquid volume,200 ml/500 ml.[Conclusion] The best medium compound and fermentation condition for production of trehalose from sweet potato starch in the small-sized trial were determined,which would provide the scientific basis for its industrial production.
