摘要
目的探讨乌梅炒炭炮制前后DNA指纹图谱特征。方法采用随机引物扩增多态性DNA(RAPD)技术扩增植物基因组DNA样品。从10个引物中筛选出能稳定扩增的引物S362(5'-GTCTCCGCAA-3')对不同产地乌梅进行扩增。结果此种分子生物学鉴别方法可以获得清晰可靠的DNA指纹图谱,不同产地的乌梅生品DNA指纹图谱一致,均有900bp、750 bp、650 bp、550 bp、300 bp、200 bp条带;经炮制后的乌梅炭DNA指纹图谱与生品有较大差异,缺少较长碱基对的扩增条带,均多出了400 bp的条带。结论 RAPD技术可作为乌梅生品道地品种的鉴定的参考方法。
AIM To study the DNA fingerprint of crude and processed Mume Fructus.METHODS DNA fingerprint of samples of crude and processed Mume Fructus were examined by RAPD.S362(5′-GTCTCCGCAA-3′) was selected from ten random primers and could be successfully used for amplifying the DNA of crude and processed Mume Fructus.RESULTS All crude Mume Fructus showed similar fingerprint characteristics of amplified six bands 900 bp,750 bp,650 bp,550 bp,300 bp and 200 bp,while the processed Mume Fructus products indicated considerable different results compared with the crude and was devoid of longer amplified band of base pairs but added band of 400 bp.CONCLUSION DNA fingerprint is a reliable method for differentiation crude Mume Fructus.
