摘要
目的观察高糖环境下豚鼠膀胱Cajal间质细胞超微结构的改变,探讨糖尿病膀胱(DCP)的发病机制。方法采用酶消化法原代培养,透射电镜观察不同葡萄糖浓度下培养的ICCs超微结构。结果随糖浓度增高和培养时间延长,ICCs内细胞器明显减少,线粒体大小不等、数量减少、肿胀、空泡样变甚至溶解,内质网扩张、空泡样变,胞质广泛溶解,突起消失。结论高糖环境对豚鼠膀胱ICCs超微结构造成显著影响,这种结构破坏可能是造成DCP发病的重要原因之一。
Objective To investigate the changes of the guinea pig bladder Cajal interstitial cells' ultrastructural in the high glucose environment,in order to explore the mechanism of diabetic cystopathy(DCP).Methods Primary culture ICCs bladder cells with collagenase digestion in vitro.Observed their ultrastructural of ICCs by transmission electron microscope at different concentrations and different times.Results The organelles in ICCs obviously diminution as the glucose concentration and culture time increased.Their mitochondrion were inequality of size,abated of quantity,swelling and distension,cavity affected and even dissolution.Endoplasmic reticulums were extension,cavity affected.Cytoplasm were deliquesced generally.Scabrosity vanished.Conclusion High glucose environment significantly affected the ICCs' ultrastructural,and these disorganization would be one of the most important etiological factor for DCP.
