摘要
目的观察CD147-shRNA重组质粒对肝癌细胞HepG2细胞MMP-9和AFP表达的影响。方法将前期合成并筛选的CD147-shRNA重组质粒pBS/U6/CD147-shRNA3转染至HepG2细胞中,采用Westernblot方法检测HepG2细胞MMP-9和AFP的表达情况。结果 HepG2细胞在转染了0.3μg和1μg的pBS/U6/CD147-shRNA3后,HepG2细胞MMP-9前体和活化型以及肿瘤标记物AFP的生成和表达明显受抑,AFP可实现80%~90%左右的抑制。结论基因沉默CD147基因对肝癌细胞HepG2的MMP-9和AFP的表达有明显抑制作用。
Objective To observe the influence of CD147-shRNA on expression of MMP-9 and AFP of HepG2 cell.Methods Transfect the pBS/U6/CD147-shRNA3 synthesized and screened before in our laboratory into HepG2 cell,detect the expression of MMP-9 and AFP by Westernblot.Results The expression of precursor and activated MMP-9 was prohibited after transfect 0.3 μg and 1 μg pBS/U6/ CD147-shRNA3;the expression of AFP was suppressed by about 80%~90%.Conclusions Silence CD147 gene by RNAi can obviously inhibit the expression of MMP-9 and AFP of HepG2 cell.
