摘要
目的:探讨血管生成素2(Ang-2)和血管内皮生长因子(VEGF)在大肠癌发生、发展、浸润、转移及血管生成中的调节作用。方法:运用免疫组织化学法分别检测85例大肠癌组织,24例癌旁肠组织,24例正常肠组织中Ang-2和VEGF的表达情况。根据CD34的表达分别计算大肠癌组织、癌旁肠组织及正常肠黏膜的微血管密度。结果:原发性大肠癌组织中Ang-2表达阳性率为63%(54/85),VEGF表达阳性率为80%(65/85),两者表达部位相似。在癌组织中,Ang-2和VEGF表达增高也可见MVD增高,但在癌旁组织中MVD不与两者表达水平平行。Ang-2、VEGF、MVD均与大肠癌组织分化程度、Dukes分期、转移密切相关。在24例癌旁组织中Ang-2和VEGF分别有10例和15例表达阳性,表达强度与癌组织无区别。在24例正常黏膜中,Ang-2几乎不表达,有10例组织呈VEGF弱表达。结论:Ang-2和VEGF均与大肠癌血管生成有关,两者协同作用,与大肠癌浸润、转移有关。Ang-2在肿瘤组织中特异性表达,可能与肿瘤的发生有关。
Objective:To analyze the expression of Ang- 2 and VEGF in colorectal cancer and their relation to angiogenesis. Methods: Tissue sections of 85 colorectal cancers were examined by immunohistochemistry for the expression of Ang- 2 and VEGF. Microvessel density (MVD) was counted by endothelial cells immunostained with CD34 antibody. Results:Ang- 2 and VEGF was detected in 63% (54/85) and 80% (65/85) of coloreetal cancer, respectively. They were expressed at similar position of the same tissue slice. The level of Ang - 2 and VEGF expression and MVD are related with the differentiation, Dukes stages and lymph metastasis. In 24 para - cancer tissues, 10 and 15 samples were immunostained by Ang - 2 and VEGF, respectively. Normal colorectal tissue is little immunostained by Ang- 2 antibody, but 10 of 25 normal coloreetal tissue was slightly immunostained by VEGF antibody. Conclusion:The expression of Ang - 2 and VEGF favor angiogenesis of colorectal cancer, and furthermore advance cancer proliferation, metastasis and invasion. Ang- 2 is specially expressed in cancer tissue and may be related with tumorigenesis.
