分泌抗人IgMμ链单克隆抗体杂交瘤细胞株的建立 被引量:1

ESTABLISHMENT OF MOUSE HYBRIDOMA CELLS SECRETING ANTI-HUMAN IgMμ CHAIN MONOCLONAL ANTIBODY

摘要 本文采用杂交瘤技术建立了6株分泌抗人IgMμ链单克隆抗体的杂交瘤细胞株.经琼脂糖双扩散试验、双扩散和ELISA封闭试验,以及免疫电泳等方法证实,所分泌的抗体具有抗人IgMμ链特异性.其中4株分泌IgG_1亚类、2株分泌IgG_2a亚类抗体.经一年多的反复冻存和复苏,这6株细胞均能稳定分泌高效价抗体. 6 strains of mouse hybridoma cells secreting anti-human IgMμ chain McAbs were successfully established with hybriotechnique. The specificity of McAbs was identified by means of double diffusion, blocking tests and immunoelectrophoresis. Of the 6 hybridoma cells, 4 secrete mouse IgG_1 and 2 IgG_(2a) subvarieties. After repeated frozen storage and thawed revival for more than one year, they are still steadily producing high titers of McAbs. The application of McAbs in research work and clinical immunodiagnosis as reagents is being studied.
