摘要
本文探讨了提高大麦黄花叶病毒机械接种发病率有关的条件和因素,试验结果表明以0.1M pH7.0磷酸盐缓冲液(含DIECA)1~5倍量,用新鲜或短期冰冻贮存的病株为接种毒源,接种于不完全叶麦苗,在18~20℃,每天光照(50001x以上)10~12h的条件卜,株发病率可达90%。本方法可试用于大麦品种抗黄花叶病的鉴定。同时用机械接种法测定病毒粗汁液的钝化温度为55~60℃,10min,稀释终点为10^(-3),体外保毒期(20℃)约2天。
The factors and conditions for increasing disease incidence of barley yellow mosaic virus (BaYMV) by mechanical inoculation were discussed. The results showed that the disease incidence was increased up to 90% or higher as the fresh infected barley leaves or shortly frozen stored ones were used as sources for the inoculation on barley seedlings at incomplete leaf stages under the condition of 18-20℃ temperature and 5000 lx or more for 10-12 hours per day. The results also presented that the thermal inactivation point (TIP) (10 min.) was 55-60℃ and the dilution end point (DEP) was 10^(-3) as well as the longevity in vitro (LIV) was about 2 days.
