摘要
以草棉(Gossypium herbaceum L.)的根尖为材料,采用5μg/ml放线菌素D(AMD)+0.002mol/l 8—羟基喹啉混合液处理1.5h,Ohnuki′s溶液中低渗0.5h。根尖在载玻片上加固定液捣碎法制片,首次在草棉中诱导出高分辨的染色体带纹。显示的带纹随染色体所处阶段而有所变化,前期和晚前期染色体带纹最多。早中期次之,中期染色体仍能显示1~4个带纹。
Root tips of Gossypium herbaceum L. were treated -with mixture of 5 μ g/ml actino-mycin D (AMD)+0.002 mol/L S-Hydroxyqinoline for 1.5 hours, then with Ohnuki's hypotonic solution for o.r?hour. The root tips ware 'put on clean slides and were smashed and smeared in metha nol acid (3: 1 ) fixative solution. High-resolution banding was induced for the first time in Gossypium. The number of bands varied with mitotic periods, with more bands shown fro.n early prophase to late prophase, and less in prometaphase. One to four bands were still distinguishable by the metaphase chromosomes.
