红藻氨酸致癫痫大鼠血清和脑脊液中S100B、CRP的动态变化 被引量:2

Study of the changing levels of S100B and C-reactive protein in serum and cerebrospinal fluid of kainic acid-induced epileptic rats

摘要 目的观察致痫大鼠血清和脑脊液中S100B、C反应蛋白(CRP)含量的动态变化,以探讨这两种生化指标与癫痫的关系。方法雄性SD大鼠48只随机分为对照组(8只)和模型组(40只),模型组再依据造模后大鼠处死时间分成6 h、12 h、24 h、72 h和1周共5组。采用红藻氨酸(KA)侧脑室注射建立癫痫动物模型,ELISA法测定大鼠造模后各时间点血清和脑脊液中S100B、CRP的含量,并与对照组比较。结果与对照组比较,模型组大鼠血清和脑脊液中S100B、CRP含量在癫痫发作后6 h逐渐增加,24 h达高峰,以后逐渐降低;血清和脑脊液中S100B、CRP水平均呈正相关(r=0.788和0.873,P均<0.01)。结论致痫大鼠脑内存在动态的急性炎症反应,这种炎症反应可能参与癫痫发生及癫痫所致的脑损伤。血清和脑脊液中S100B、CRP可用于检测这种炎症反应和脑组织损伤。 Objective To investigate the levels of S100B and C-reactive protein(CRP) in serum and cerebrospinal fluid(CSF) of epileptic rats,and then to approach the relationship between the two biochemical indicators with epilepsy.Methods 48 adult male SD rats were randomly divided into control group(n=8) and model group(n=40).According to the time when rats were killed after the success of model making,model group was divided into five groups(6 h,12 h,24 h,72 h and 1 w).Animal model of epilepsy was established in rats by intracerebroventricular injection of kainic acid(KA).Levels of S100B and CRP in serum and CSF within various phases after seizures were measured by ELISA and contrasted with control group.Results Compared with control group,levels of S100B and CRP in serum and CSF of model groups increased gradually at 6 h after seizures and declined gradually after reaching the peak at 24 h.Both in serum and CSF,levels of S100B were positive correlation with CRP(r=0.788 and 0.873 respectively,P0.01).Conclusions Inside the brain tissues of epileptic rats,there are changing acute inflammatory reactions which maybe involve in epilepsy and seizure-induced brain damage.Both of S100B and CRP in serum and CSF may be used to predict the inflammatory reaction and brain damage in epileptic rats.
