嗜酸乳杆菌部分生理特性的研究

摘要 目的:对嗜酸乳杆菌生长曲线等生理特性进行研究,初步掌握该菌的生长、繁殖和代谢规律。方法:按国标GB/T4789.35-2003的方法测定菌落总数。以培养时间为横坐标,pH值、滴定酸度和菌落总数对数为纵坐标,绘制菌株(种)生长曲线。通过测定pH值及OD值,确定菌株(种)的最佳培养条件。通过测定活菌数,确定菌株(种)的接种方法及生长促进因子。结果:综合分析生长曲线,确定嗜酸乳杆菌在5%接种量,37℃下最佳收获期为12h~16h;最佳培养条件:初始pH值5.8,37℃培养;选择4种天然廉价物料(添加剂)作为菌株(种)的促生因子,发现对其生长均有促进作用,影响大小依次为:西红柿汁5%>香菇汁3%>胡萝卜汁5%>啤酒5%>对照。结论:得到嗜酸乳杆菌的最佳生长条件,为菌液培养、冻干粉制备及活菌制剂打下基础。 Objective:Study of physiological property of Lactobacillus acidophilus about growth curve istobe rwcwe of the regulatiem of its grouth,proliferation and metabotism.Methods:the total number of colonies was determined according to national standard GB / T 4789.35 -2003.Set incubation time as the abscissas、PH、titratable acidity and the total number of colonies as ordinate.Draw the growth curve of bacterial strain.The best condition of culture of two bacterial strains was determined by measured PH and OD value. While,inoculation method and growth - promoting factor were determined through measured viable count.Results:To analyzed growth curve,the best harvest of Lactobacillus acidophilus was 12 - 16 hours under the condition of 5%inoculum size,37℃.Its best condition of culture was pH =5.8,37℃;there is facilitation of growth to select four low - cost natural materials(additives),and effect from strong to weak is tomato juice,5% mushroom juice,3% 5%carrot juice Beer 5% control group.Conclusion:The best condition of culture of Lactobacillus acidophilus,lays foundations for cultivation broth,freeze - dried powder preparation and viable bacteria preparation.
