摘要
O-聚糖(O-glycan)的硫酸化是指在硫转移酶的作用下,将硫酸根成分从3'-磷酸腺苷5'-磷酸硫酸盐转移至O-聚糖的羟基基团上。硫酸化的O-聚糖是表达于淋巴结高内皮小静脉(HEV)上的L-选凝素配体的重要成分,O-聚糖的硫酸化在淋巴细胞的再循环及炎症中均发挥关键作用。内皮细胞上的硫转移酶对O-聚糖的硫酸化起着决定性的调节作用。慢性炎症反应中内皮性硫转移酶的表达显著增加。据此,可用各种动物模型对硫酸化为O-聚糖及硫转移酶与炎症的关系作进一步研究;可以制备相应的O-聚糖硫酸盐的特异抗体,寻找重要的广谱抗炎治疗靶点;还可以通过调节硫转移酶的表达来控制O-聚糖的硫酸化,从而实现对炎症的有效治疗。本文对免疫学上其它重要的硫酸化表位也进行了阐述。
Sulfation of O-glycan involves transfer of a sulfate moiety from 3'-phosphoadenosine 5'-phosphosulfate (PAPS) to hydroxyl group by sulfotransferases (STs). Sulfated O-glycan, an important element of L-selectin ligand expressing on the high endothelial venules (HEVs) of lymphoid nodes, plays a key role in both lymphocyte recirculation and inflammation. Endothelial STs regulate the sulfation of O-glycan. And the expression of endothelial STs is increased significantly in chronic inflammation. Therefore, further researches involving the relationships between either sulfated O-glycan or STs and inflammation ought to be carried out in various animal models, and selecting specific antibodies against O-glycan sulfate is able to discover the new target to cure inflammation broad-spectrumly. Furthermore, the effective treatment of imflammation could be achieved by regulating the level of STs which control the sulfation of O-glycan. Other lmmunologically important sulfotopes are also discussed in this review.
