摘要 目的:建立以艾司唑仑为内标的高效液相色谱法测定人血浆中拉莫三嗪浓度的方法。方法:色谱柱为HyPersil ODS2,流动相为乙腈:0.05%磷酸:36:64;流速为1.0mL·min-1,柱温为35℃,检测波长为240nm,进样量为20pL。结果:拉莫三嗪血药浓度在0.95~38μg·mL-1。范围内线性关系良好(r=0.9991);方法回收率为99.77%-104.32%;日内、日间RSD分别为1.35%~2.07%、3.04%~6.82%。结论:本方法操作简便、快速、灵敏、重复性好、回收率及精密度高,可用于拉莫三嗪的治疗药物监测。 Objective:To determine the lamotrigine level in the plasma of children with epilepsy by RP-- HPLC. Methods: The chromatographic column was Hypersil ODS2 (250mm× 4.6mm, 5μm) column with a mobile phase consisting of methanol--0.05% phosphoric acid solution(36 : 64). The flow rate was set at 1. 0mL.min-1 and the detection wavelength was at 240nm. The column temperature was room temperature and the injection volume was 20μL. Results: The linear range of lamotrigine was from 0. 95 to 38μg·mL-1 (r = 0. 9991) with the recovery of 99.77%-104.32%. The RSD of intra--day and inter--day was 1.35%- 2.07% and 3.04%- 6. 82%, respectively. Conclusion: The method is simple, rapid, sensitive and reproducible with high recovery and good precision for pharmaceutical monitoring of larnotrigine.
