竹种内转录间隔区的核苷酸(ITS)序列分析

摘要 以慈竹、龙竹和紫竹3个竹种为研究对象,通过对竹子的新鲜叶片中的rDNA的ITS序列进行聚合酶PCR扩增,以及对其进行序列测定及分析。检测出目的片段ITS总长分别为慈竹390 bp、龙竹384 bp、紫竹380 bp。3种不同属的竹种之间的ITS序列有差异。同时在国家生物多样性保护信息系统(NCBI)上选取同属竹种ITS序列进行比较分析。以此建立系统树,用以分析它们之间的亲缘进化关系。 Using the fresh leaves of Neosinocalamus affinis,Dendrocalamus giganteus,Phyllostachys nigra as research materials,the extraction of DNA was done,and the genes of Internal Transcribed Spacer were amplified from genomic DNA by polymerase-chain-reaction(PCR).Then the PCR products were sequenced and analyzed.The lengths of the PCR production were 390 bp,384 bp,and 380 bp for N.affinis,D.giganteus,and Phy.nigra respectively.The sequences are different to each other.Meantime,other ITS sequences of the genus in the NCBI were selected.Then the phylogenetic tree was established on the basis of the ITS sequences in order to analyze their evolutionary relationships.
