摘要
利用灭活的猪血凝性脑脊髓炎病毒加弗氏完全佐剂和弗氏不完全佐剂免疫家兔,制备高免血清,测定其效价;用硫酸铵-蛋白A亲和层析法分离纯化兔血清IgG,并采用SDS-PAGE和ELISA对IgG纯化产物进行检测。结果表明,兔抗HEV高免血清的效价为1:12800;采血并分离血清,经过亲和层析纯化得到的兔抗HEVIgG,效价达150000。
In this study, hyperimnmne serum was prepared to inactivale porcine hemagglutinating encephalomyelitis virus with Freund＇s complete adjuvant and Freund＇ s incomplete adjuvant in rabbits; then rabbit serum IgG was purified by ammonium sulfateprotein A affinity chromatography. The effct of purified lgG was tested by SDS-PAGE and ELISA. The results showed that the titer of rabbit hyperimmune serum anti-HEV was 1：12 800; the titer of IgG anti-HEV was 1：50 000.
