摘要
目的研究中药黄芪(AMB)提取物对体外培养的人肝癌细胞的抑癌活性。方法 AMB经粉碎、浸提、超速离心、减压浓缩、冷冻干燥得AMB提取物。人肝癌SMMC-7721细胞经AMB提取物处理不同时间后以MTT法测其对人肝癌SMMC-7721细胞线粒体代谢活性的影响,以细胞记数法观察了其对细胞增殖的影响。结果①AMB提取物明显抑制人肝癌SMMC-7721细胞的生长,降低线粒体代谢活性;②经AMB提取物处理24h后的人肝癌细胞,即使更换为正常培养基,其线粒体代谢活性及细胞增殖与对照相比仍差异显著。结论中药AMB提取物含抑癌活性物质,能抑制体外培养的肝癌细胞增殖和降低线粒体代谢活性。
AIM To demonstrate the effects of the extracts from Chinese traditional medicine Astragalus membranaceus (Fisch). Bge Var. mongholicus (Beg) Hsiao (AMB) on the proliferation and mitochondria metabolism of human hepatoma SMMC-7721 cells.METHODS The dried AMB was broken to powder, extracted in cold DW and centrifuged to discard the pellet and the AMB supernatant was concentrated and lyophilized. After the test of dose dependent experiments, human hepatoma cells were treated with 700 mg L AMB extracts for 48 h and mitochondria metabolic activity was detected by MTT method. The effects of AMB extracts on the proliferation were observed with the treatment of 700 mg/ L AMB extracts for 7 d and the proliferation of human hepatoma was also examined after treatment with AMB extracts for 24 h and then replaced by culture in normal media (containing 100 mL/L FCS) for 96 h.RESULTS The lowest efficient dose was about 400 mg/ L AMB extracts. AMB extracts could significantly decrease the activity of mitochondria metabolism. The A570 absorption in AMB treatment group was only 50.9% of that in control group after 48 h treatment and 44.1% after 72 h treatment. During the first 48 h treatment, the cell number maintained the level of that before treatment, after 48h the cell number was gradually decreased and showed great differences as compared with the control groups (only 15.9% of control group) while it did not increase within 48 h in recover culture alter treatment with AMB extracts for 24 h, and after 48 h the cells went into logarithmic phase.CONCLUSION AMB extracts could inhibit the proliferation and mitochondria metabolic activity of human hepatoma cells.
