摘要
目的探讨粒细胞集落刺激因子(G-CSF)对脑挫伤大鼠血浆及脑组织中血管紧张素Ⅱ(AngⅡ)和内皮素(ET)含量的影响.方法采用自由落体法致SD大鼠脑挫伤动物模型.将大鼠随机分为治疗组、对照组和正常组,治疗组脑挫伤后2h开始应用G-CSF(浓度为2mg/L)皮下注射10μg/kg/d,共5d.对照组和正常组2h后给予等量的生理盐水皮下注射,共5d.正常组假手术不致伤.每组大鼠再随机分为两个亚组分别为治疗1周组、治疗2周组、对照1周组、对照2周组、正常1周组和正常2周组.分别在造模成功1周和2周后检测各组血浆及脑组织中AngⅡ和ET的含量.结果 1周:对照组血浆及脑组织中AngⅡ和ET的含量均增高,与正常组比较有差异(P<0.05).2周:对照组血浆及脑组织中AngⅡ和ET的含量增加更加明显,与正常组比较有显著的差异(P<0.01).治疗组1周和2周的结果是血浆及脑组织中AngⅡ和ET的含量与对照组比较均有降低,差异有统计学意义(P<0.05).结论脑挫伤大鼠血浆及脑组织中AngⅡ和ET的含量明显升高,G-CSF能降低两者的含量,从而起到对受损伤的脑组织的保护作用.
Objective To investigate the influence of granulocyte colony stimulating factor (G-CSF) on angiotensin Ⅱ (AngⅡ) and endothelin (ET) content in plasma and brain tissue of brain contusion rats. Methods SD rat models of brain contusion established by free fall method were divided ramdomly into 3 groups including treatment group, control group and normal group. Injected rhG-CSF which was diluted with normal saline (the concentration is 2 mg/L) 10 ug/kg/d after treatment group model had been completed 2 hours for 5 days. Rats of control group and normal group were injected the same amount of saline 2 hours later for 5days. Rats of Normal group were not hurt. Rats of each group were randomly divided into two subgroups: 1 W, 2 W treatment groups, 1 W, 2 W control groups, 1 W and 2 W normal groups. The content of Ang Ⅱ and ET in plasma and brain tissue of every group was tested after completing the model one and two weeks. Results In comparison with that of normal group, Ang Ⅱ and ET were both increased significantly in control group after one week ( P 〈0.05). After 2 weeks AngⅡ and ET were both increased more significantly in control group in comparison with that of normal group ( P 〈 0.01). Compared with that of control group, Ang lI and ET in the plasma and brain tissue decreased significantly in treatment group ( P 〈0.05). Conclusion G-CSF can increase the content of Ang Ⅱ and ET in brain contusion rat plasma and brain tissue, G-CSF has a protective effect on the injured brain tissue.
