摘要
目的:研究苦杏仁苷对大鼠肝星状细胞HSC-T6增殖的影响和对凋亡基因Bax的调控作用。方法:利用噻唑蓝染色法动态检测不同浓度的苦杏仁苷对大鼠HSC-T6细胞增殖的影响,利用PCR法检测苦杏仁苷对HSC-T6细胞凋亡基因Bax的表达水平。结果:苦杏仁各实验浓度组对HSC-T6细胞的增殖无直接抑制作用,但其高浓度10-3mol/L和中浓度10-4mol/L可以使HSC-T6细胞提前到达增殖高峰期;而各浓度组对HSC-T6细胞的Bax基因都有一定的诱导作用,并且存在量效关系。结论:苦杏仁苷不能直接抑制HSC-T6细胞的增殖,但可能通过诱导或上调凋亡基因Bax的过度表达,使细胞提前进入凋亡程序阶段,最终使活化的HSC-T6发生细胞凋亡,以起到抗肝纤维化的作用。
Objective: To observe the effect of amygdalin on the proliferation of HSC-T6 and Bax gene expression. Methods. Proliferation of HSC-T6 cultured in the medium containing different concentrations of amygdalin was dynamically detected by methyl thiazolyl tetrazolium (M-I-F) assat. Bax gene expression level was detected by polymerase chain reaction (PCR) technique. Results: No direct inhibition effect was shown on the proliferation of HSC-T6 in all of amygdalin groups. But in amygdalin groups at 10-3 mol/L and 10-4mol/L, HSC-T6 reached the growth peak in advance, Amygdalin had certain effect on inducing Bax gene expression, and .the effect was in dose-dependent manner. Conclusion: Amygdalin has no direct inhibition effect on proliferation of HSC-T6, but may induce or up-regulate Bax gene expression to promote apoptosis for anti-hepatic fibrosis,
