摘要
目的探讨人参皂甙Rb1(Gs-Rb1)是否可减轻阿霉素所致的心力衰竭以及这种效应是否与减轻心肌细胞凋亡有关。方法阿霉素诱导慢性心力衰竭模型完成后,心力衰竭大鼠被随机分为Gs-Rb1组17只(70mg.kg-1.d-1)和对照组15只。第4周检测心脏超声、左心室脑利钠肽(BNP)、心肌肌钙蛋白Ⅰ(cTnⅠ)和原位缺口末端标记法(TUNEL)。结果①与对照组相比,Gs-Rb1组左室射血分数(LVEF,P=0.015)和左室短轴缩短率(LVFS,P=0.022)显著改善,全心质量指数(HW/BW,P=0.005)及左室质量指数(LW/BW,P=0.000)均显著改善;②Gs-Rb1组的BNP水平(P=0.000)和cTnⅠ水平(P=0.000)均显著低于对照组;③与对照组相比,Gs-Rb1组的心肌细胞凋亡率显著下降(P=0.045);④LVEF和心肌cTnⅠ水平(r=-0.55,P=0.045)、心肌细胞凋亡率(r=-0.44,P=0.025)呈负相关,心肌cTnⅠ水平和心肌细胞凋亡率呈正相关(r=0.51,P=0.016)。结论人参皂甙Rbl可改善阿霉素所致的心力衰竭,此效应可能与减轻心肌细胞凋亡有关。
Objective On the basis of adriamycin-induced chronic heart failure (CHF), the present study aimed at investigating whether Ginsenosides-Rbl (Gs-Rbl) improved cardiac function and whether Gs-Rbl relieved the apoptosis of cardiomyocytes in vivo. Methods CHF models of rats were established by adriamycin, rats were randomly divided into control group (n^15) and Gs-Rbl group (70 mg ~ kg ~ ~ d ~ , n--17). At the 4th week, echocardiography examinina- tion, brain natriuretic peptide (BNP), Cardiac troponin I (eTnI) and TdT--mediated dUTP nick end labeling (TUNEI.) were analyzed. Results @Compared to control group, Gs-Rbl significantly improved left ventricular ejection fraction (LVEF, P=0. 015) and left ventricular fraetional shortening (LVFS, P=0. 022) ; @Gs-Rbl significantly degraded the concentration of BNP (P=0. 000) and cTn I(P=0. 000) in blood; (~)The apoptosis ratio of eardiomyocyte was down- regulated in Gs-Rbl group than that in control group (P= 0. 045) ; @There was a significant negative correlation between LVEF and cTn I (r:--0.55, P=0. 045), between LVEF and the apoptosis ratio (r=--0.44, P=0. 025) ; a signifi- cant positive correlation between the apoptosis ratio and cTn I (r= 0. 51, P= 0. 016). Conclusion Gs-Rbl may improve cardiac function in CHF rats induced by adriamyein, in which the effects of Gs-Rbl relieving the apoptosis of cardiomyo cytes may play a key role.
