摘要
以乙酸乙酯为萃取剂,虫草素得率与纯度为考察指标,在研究粗粉液浓度、单位体积粗粉液的乙酸乙酯用量、摇匀时间、静置萃取时间、萃取温度、粗粉液pH值、萃取次数等单因子对虫草素萃取效果影响的基础上,通过正交试验与验证试验,优化筛选了乙酸乙酯萃取虫草素的工艺参数。结果表明,当主要考虑虫草素得率时,最佳萃取参数为粗粉液质量分数为20%、粗粉液与乙酸乙酯体积比为1:1.2、粗粉液pH为7.5、摇匀时间55min、静置萃取时间80min、26℃—28℃,萃取1、2、3次的虫草素得率分别为51%、63%、80%,纯度分别为76%、69%、58%;当主要考虑虫草素纯度时,最佳萃取参数为粗粉液质量分数为25%、粗粉液与乙酸乙酯体积比为l:1.2、pH为7.5、摇匀时间55 min、静置萃取时间90min、26℃—28℃,萃取l、2、3次的虫草素得率依次为42%、67%、77%,纯度依次为78%、72%、67%。与当前虫草素经典提取,纯化工艺相比,该工艺具有较好的实用性、针对性和参考价值。
Based on the study of verification test and via orthogonal test in extracting cordycepin of different factors including concentration of cordycepin infusion,dosage of ethyl acetate,extracting standing time,shaking time,extracting temperature,pH value of cordycepin infusion and extractingtimes,the best processing parameters for extracting cordycepin were optimized and screened.The resultsshowed that when mainly considering the yield rate of cordycepin,the best parameters were as follows:massfraction of cordycepin infusion of 20%,cordycepin infusion to ethyl acetate ratio of 1:1.2,pH of cordlycepin infusion of 7.5,shaking time of 55min,standing time of 80 min,and extracting temperature of 26℃-28℃.Under the optimal conditions,the yield rate and purity of cordycepin were 51%and 76% for one extracting time,63%and 69%fortwo extracting times,and 80%and 58%for 3 extracting times,respectively.When mainly considering the purity of cordycepin,the best parameters were as follows:mass fraction of cordycepin infusion of 25%,cordycepin infusion to ethyl acetate ratio of 1:1.2,pH of 7.5,shaking time of 55 min,standing time of 90 min,and extracting temperature of 26℃-28℃.the yield rate and purity of cordycepin were 42%and 78%for one extracting time,67%and 72%for two extracting times,and 77%and 67%for three extracting times,respectively.Comparing with the current extraction process of cordycepin by ethyl acetate,the optimized process hadbetter practical and reference value.
