摘要
目的应用反相高效液相色谱法(RP-HPLC)测定羊水和胎盘中的乳酸依沙吖啶。方法 采用甲醇浸泡溶解,7%三氯乙酸沉淀蛋白的方法,提取样品后进行高效液相色谱测定。色谱柱:用Acclaim C18色谱柱(250mm×4.6mm,5μm);流动相:甲醇-乙腈-水(50∶28∶22);紫外检测器:检测波长:272nm;柱温:35℃;流速:0.8mL·min-1。结果乳酸依沙吖啶在1.0～10.0μg·mL-1与峰面积呈现良好线性关系(r=0.999 9),回收率为95.5%～99.5%,日内RSD为1.2%～1.7%,日间RSD为1.7%～2.2%。结论该方法简便、快速、灵敏、准确,可作为临床检测方法。
Objective To establish an HPLC method to the monitor ethacridine lactate in the placenta and amniotic flu id. Methods The sample was dissolved in the methanol and 7% trichloroacetic acid （TDA） was used to subside the protein. The column was acclaim C18 （250 mm×4.6 mm, 5 μm）, the mobile phase was methanol-acetonitrile-water （50 ： 28 ： 22）, the wavelength was 272 nm, the column temperature was 35 ℃, and the flow rate was 0. 8 mL · min^-1. Results At 1.0-10.0 μg · mL^-1, a good linearity （r=0. 999 9） was shown. The recovery was 95.5%- 99.5%, and intra-dayRSD was 1.2%-1.7%, and inter-day RSD was 1.7%-2.2%. Conclusion The method is simple, rapid, sensitive, and can be used in clinical tests.
