摘要
目的建立高效液相色谱法测定人血浆中硫酸头孢匹罗的含量。方法采用Diamonsil C18色谱柱,流动相为0.03mol.L-1磷酸二氢铵缓冲液(pH 3.3)-乙腈(9∶1);流量为1.0mL.min-1,检测波长为270nm。结果硫酸头孢匹罗浓度在0.3~20.0μg.mL-1呈良好的线性关系,最低检测浓度为100ng.mL-1,日间、日内精密度均<6.7%,回收率均>80%。结论本方法简单快速,准确灵敏,适用于注射用硫酸头孢匹罗在人体内的药物动力学研究。
Objective To establish an HPLC method for the determination of cefpirome sulfate in human plasma.Methods Diamonsil C18 colume was used with 0.03 mol·L-1 ammonium dihydrogen phosphate buffer fluid-acetonitrile(9∶1) as the mobile phase at 1.0 mL·min-1.The detection wavelength was 270 nm.Results Cefpirome sulfate had good linear relationship in 0.3-20.0 μg·mL-1.The minimum detectable concentration was 100 ng·mL-1.The variations for intra-and inter-day precision were less than 6.7%,and the extraction recoveries were more than 80%.Conclusion The method is simple,sensitive and accurate.It is suitable for the pharmacokinetics of cefpirome sulfate in human plasma.
