摘要
目的探讨肿瘤坏死因子基因6个SNPs位点多态性与河南汉族原发性高血压(EH)的相关性。方法采用限制性扩增片段长度多态性(PCR-RFLP)的方法检测TNF-α启动区基因-238G>A、-308G>A、-850C>T、-857C>T、-863C>A及TNF-β第一内含子基因+252G>A多态性,并运用卡方检验探讨各SNP位点多态性与EH的相关性。结果 TNF-α-308G>A位点基因型分布和等位基因频率在人群中存在差异显著性(χ2=6.407,P=0.041;χ2=4.781,P=0.029),EH组中,TNF-α-308G>A中基因型GA+AA人群的收缩压(SBP)、舒张压(DBP)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、尿酸(UA)及空腹血糖(FPG)含量明显高于GG基因型人群(P<0.05);TNF-α基因-238G>A、-850C>T、-857C>T、-863C>A及TNF-β基因+252G>A多态性各基因型及等位基因频率在EH组和对照组的分布差异无统计学意义。结论 TNF-α基因-308G>A位点基因多态性与河南汉族原发性高血压的发病相关。TNF-α-238G>A、-850C>T、-857C>T、-863C>A及TNF-β+252G>A位点基因多态性与河南汉族原发性高血压无关联。
Objective To investigate the correlation between 6 SNPs on tumor necrosis factor gene and essential hypertension (EH) in Han Chinese in Henan province. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to evaluate the gene polymorphisms including TNF-α-238G〉A, -308G〉A, -850C〉T, -857C 〉T, -863C 〉A gene and TNF-β +252G〉A gene in 260 unrelated EH patients (EH group) and 394 healthy population (control group). Results The frequencies of the GA +AA genotype and allele of TNF-α-308G 〉A in EH patients were significantly higher than those in heahh control group (x2=6.407andx2=4.781,allP〈0.05),inEHpatients, theSBP, DBP, HDL, LDL, UA and FGB values were significant difference between the GA+AA genotype and GG genotype subgroups (P〈0.05). There was no significant difference in genotype and allelic frequency of the TNF-α-238G〉A, -850C〉T, -857C〉T, -863C〉A gene and TNF-β +252G 〉A polymorphism between healthy subjects and EH patients. Conclusion The TNF-α-308G 〉A gene polymorphism is positively eorrelated with EH of Han Chinese in Henan. And the TNF-α-238G〉A, -850C〉T, -857C〉T, -863C〉A gene and TNF-β+252G〉A gene polymorphism is not associated with the susceptibility to EH.
