摘要
在5L发酵罐水平上研究不同培养条件对富硒产朊假丝酵母细胞生长、谷胱甘肽合成和富硒能力的影响,通过分批发酵试验确定较优的培养方案:在发酵培养的第18小时加入20mg/L亚硒酸钠,发酵罐转速450r/min,L-蛋氨酸的添加浓度10 mmol/L。该条件下,酵母细胞干重为11.32g/L,胞内谷胱甘肽和有机硒含量分别达到11.5mg/g和1 352μg/g,该方案为高性能(高胞内谷胱甘肽含量、高有机硒含量)富硒产朊假丝酵母的高效制备奠定了基础。
Studied the effects of different culture conditions on cell growth of selenium-enriched Candida utilis,the ability of glutathione biosynthesis and selenium enrichment cultured in a 5 L stirred fermentor.An optimum combination of culture conditions which involved 20 mg/L sodium selenite added to the culture medium at 18 h,together with agitation rate of 450 r/min and 10 mmol/L L-methionine added at 10h,were achieved under batch fermentation with C.utilis SZU 07-01.Under these optimal conditions,the dry cell weight was 11.32 g/L,and the contents of intracellular glutathione and organic selenium were 11.5 mg/g,1 352 μg/g,respectively.This study laid a foundation for the efficient preparation of high quality(high intracellular glutathione and high organic selenium content) selenium-enriched Candida utilis.
