摘要 目的:探讨HCV核心区基因疫苗在Balbc 小鼠的免疫应答。方法:将与HCV核心区基因互补的cDNA定向克隆于真核表达载体pcDNA3(pcDNAHCV) ,免疫Balbc 小鼠,观测小鼠血清抗HCV抗体及小鼠脾细胞对HCV 核心抗原的特异性增殖反应。结果:5 只经皮下注射的免疫鼠中有4 只出现抗HCV 抗体,经肌肉注射的免疫鼠全部出现抗体阳转,而对照组全阴性,肌肉注射免疫组抗体水平高于皮下注射免疫组,动态观察发现追加免疫2 w 后抗体即可阳转,4 ~8 w 达高峰,可持续12w 以上。免疫组小鼠脾细胞对HCV 核心抗原的刺激指数明显高于对照组。结论:pcDNAHCV在Balbc 小鼠不仅可诱导体液免疫应答,而且可诱导细胞免疫应答。 Objective:DNA basedimmunization has been provedto be an effective means of vaccination in animal models and may be promising candidates asantiviralagents.Thereisa need to develope new antiviralagentsin an attemptto eradicate persistent HCVinfection from liver.Inthisstudy,theimmuneresponsesto HCVcoregene DNA bassedimmunizationin Balbc mice wereevaluated.Methods:The HCV core genecDNA(genotype:II)wasinsertedintothe eukaryoticexpression vectorunderthe CMVpromoterbetween EcoRIand XbalIsite(pcD NA HCV) .The Balbc mice wereimmunized by multiplesitesintramuscularorsubcutantousinjection with pcDNA HCV.The anti HCVAbin serum weredetected by ELISA,meanwhile,theproliferativereponsesofspleencytesto HCVcore antigens were measured by MTTmethod.Re sults:Theanti HCVAb were detectedin 4 outof5immunized bysubcutaneousinjectionandinallmiceimmunized byintramuscularinjection. Peektiter werereached by4~6 weeksafterimmunization and were maintainedforatlease3 months.Thestimulationindexofspleencytesfrom miceimmunized with pcDNA HCVto HCVcore antigen weresignificantly higherthanthatof miceinjected with pcDNA3( P< 0.01) .Con clusion:Thestudydemonstratedthatbothhumoraland cellularimmuneresponsescould beelicitedin micebypcDNA HCV.Geneimmunization can aidthe developmentof HCVvaccines.The pcDNA HCVmightbe promising candidates asantiviralagents.
