清热化瘀方对缺氧损伤神经干细胞BDNF、GDNF表达的影响

Effect of Qingre Huayu Prescription on the Expression of Brain-derived Neurotrophic Factor and Glial Cell Line-derived Neurotrophic Fator Protein in Hypoxic Neural Stem Cells

摘要 目的:观察大鼠缺氧损伤神经干细胞内BDNF、GDNF蛋白表达的变化以及清热化瘀方对其表达的影响。方法:建立缺氧损伤神经干细胞模型并制备清热化瘀方血清,应用免疫细胞化学技术检测BDNF、GDNF蛋白在缺氧损伤神经干细胞中的表达变化。结果:缺氧损伤后神经干细胞内出现BDNF、GDNF表达,清热化瘀方血清干预后能够促进二者的表达(P<0.05或0.01)。结论:清热化瘀方血清可上调GDNF、BDNF在缺氧损伤神经干细胞内的表达。 Objective:To observe expression of brain-derived neurotrophic factor(BDNF) and glial cell line-derived neurotrophic fator(GDNF) protein in hypoxic neural stem cells(NSC) of rats and effect of Qingre Huayu Prescription(QRHYP) on them.Methods:The hypoxic neural stem cells models were established by culturing the NSC at anaerobic incubator for 6h and the QRHYP serum have been separated from blood of rat.The expression change of BDNF and GDNF in NSC by immunocytochemistry technique.Results:The expression of BDNF and GDNF had not been found in normal NSC,but appeared in hypoxia NSC.And the expression level of BDNF and GDNF protein in NSC treated with QRHYP serum were higher than that in NSC treated with normal serum(P0.05 or 0.01).Conclusion:After hypoxia,the expression of BDNF and GDNF protein in NSC increased,and NYA could upregulate the expression of BDNF and GDNF.
