摘要
目的:对熏烟叠加气管内滴注脂多糖(LPS)所致慢性阻塞性肺病(COPD)急性期大鼠模型进行中医基本证候的判定。方法:通过动态采集模型动物呼吸、活动、皮毛、二便等宏观体征;运用病理形态学观察模型动物肺及支气管组织的微观变化。以临床证候诊断标准为依据,根据动物宏观体征、病理学变化等信息,通过与临床COPD各证候诊断标准进行同质性比较与鉴别,进而对动物模型进行证候属性的判定。结果:模型动物出现呼吸喘促,咳嗽,口鼻气管内分泌物明显增多,大便干结,小便颜色由淡黄色变为亮黄色,被毛灰黄无光泽,头部毛发脱落稀疏,饮食明显减少,体重增长明显减慢,舌体及爪甲颜色均较对照组深。结论:通过致病之因、模型动物的证候体征特点以及与临床各证型的比较与鉴别可知采用熏烟叠加气管内滴注LPS复合法构建COPD急性期模型与临床上COPD"痰热壅肺证"证候特点最为接近。
Objective:To investigate the syndrome property of chronic obstructive pulmonary disease(COPD) rat models by passive cigarette smoking plus intratracheal instillation of lipopolysacchride(LPS).Methods:Global physical signs observation of breathing,activity,coat,stool and other macro-signs and the images collection of the tongue and jaw were conducted dynamically at various time points after modeling.The micro-changes of lung and bronchial tissues were observed by using the methods of pathologic morphology.The syndrome property of animal model was determined by using comparison and identification of homogeneity between the information of animals and the clinical diagnosis criteria of the syndrome.Results:The rats in model group manifested dyspnea with rapid and short breath,frequent cough,nose and mouth of the tracheal secretions increased significantly,dry stool,urine color changed from light yellow to bright yellow,sallow dull coat,thin head hair loss,diet significantly reduced weight gain slowed down,the color of tongue and jaw was depth than the control group.Conclusion:By the result of disease,signs of the syndrome animal models and clinical characteristics and comparison of the various syndromes shows that rat model with passive cigarette smoking plus intratracheal instillation of lipopolysacchride(LPS) was constructed with the"phlegm obstruct the lung"syndromes of acute COPD clinical at the closest.
