摘要
目的探讨白藜芦醇对Aβ25-35诱导PC12细胞凋亡和线粒体跨膜电位的影响。方法 20μmol/LAβ25-35刺激PC12细胞后分别给予160,80,40,20μg/L,CCK-8法检测细胞活力,AnnexinV-FITC流式细胞仪检测细胞凋亡率,荧光显微镜观察细胞形态,罗丹明染色后测线粒体跨膜电位。结果与模型组比较,白藜芦醇呈剂量依赖性的提高Aβ25-35诱导的PC12细胞存活率和线粒体跨膜电位,降低PC12细胞凋亡率,有统计学差异(P<0.05)。结论白藜芦醇对Aβ25-35诱导的PC12细胞凋亡有保护效应。
AIM To investigate effect of resvertrol on apoptosis and mitochondial transmembrance potential(MTP) of PC12 cells induced by Aβ25-35.METHODS 160,80,40,20 μg/L resveratrol were adminstrated after PC12 cells induced by 20μmol/L Aβ25-35,cell viability was observed by CCK-8,apoptosis was tested by the flow cytometry,cell morphology and MTP were observed by fluorescence microscopy.RESULTS Compared with model group,resvertrol could increased the cell viability and MTP,and decreased the apoptosis in dose dependend mode.CONCLUSION Resvertrol could be against the apoptosis of PC12 cell induced by Aβ25-35.
