摘要
目的:确定HPLC法测定橄榄苦苷含量时所需的内标物及较优色谱条件。方法:用紫外分光光度计对橄榄苦苷、黄芩苷、没食子酸、绿原酸等标准物质进行全波长扫描确定检测波长,采用正交实验方法设计实验,在不同色谱条件下,测定各标准物质的保留时间、峰宽,计算分离度,根据分离度及保留时间,确定最优色谱条件及内标物。结果:正交实验结果,色谱柱不同水平间的F=39.50,流动相不同水平间的F=48.19,均>F0.05(2,2)=19(P<0.05),差异具有统计学意义,不同标准品间比较差异无统计学意义。结论:在本正交设计中,橄榄苦苷最优色谱条件为色谱柱:Agilent ZORBAX Eclipse XDB-C18(4.6×150mm,5μm),流动相:水-甲醇-甲酸为60:40:1,内标物为黄芩苷。
Objective:To determine the internal standard and chromatographic condition in determination of oleuropein content by HPLC.Methods:The ultraviolet spectrophotometer was used to scan oleutopein, baicalin, gallic acid, chlomgenic acid with full wavelength for detetmining detection wavelength. Orthogonal design experiment was appfied to determine the best chromatographic condition and internal standard by detecting retention time, peak width and calculating resolution.Results:F=39.50 of column at different levels, F=48.19 of mobile phase at different levels, which were all〉 F0.05(2,2)=19 (P〈0.05). There had no significant difference between different standard substances. Conclusions:In this orthogonal design, the best chromatographic condition of oleuropein: chromatographic column is Agilent ZORBAX Eclipse XDB-C18 (4.6 150mm, 5μm), mobile phase is water-methanol-formic acid (60:40:1), internal standard is baicalin.
