1,25(OH)_2D_3对卵巢癌细胞HO8910生长的抑制作用

摘要 为探讨1,25-二羟维生素D3[1,25-dihydroxy vitamin D3,1,25(OH)2D3]对人卵巢癌细胞HO8910生长和增殖的作用,选用人卵巢癌细胞HO8910进行体外培养。培养时添加不同浓度的1,25-二羟维生素D3,采用MTT比色法测定细胞生长抑制率,采用流式细胞术(flow cytometry,FCM)进行细胞周期及细胞凋亡分析。结果:不同浓度的1,25-二羟维生素D3对HO8910细胞生长均起到不同程度的抑制作用(P<0.05),并呈浓度和时间依赖关系;能使细胞周期时相发生改变,G1期细胞增多(P<O.05),同时降低S、G2/M期细胞比例,从而降低细胞增殖指数PI﹡(P<0.01);能够诱导细胞产生细胞凋亡(P<0.05),进而抑制卵巢癌细胞增殖。说明1,25-二羟维生素D3能抑制人卵巢癌细胞株HO8910生长;诱导细胞周期发生G1期阻滞;产生细胞凋亡作用,从而抑制癌细胞增殖。 To study the effect of 1,25-dihydroxy vitamin D3 [1,25(OH)2D3]in the proliferation of human ovarian cancer cells,HO8910 human ovarian cancer cell line were cultured in vitro and treaed with different concentrations of 1,25(OH) 2D3 for 1,2 and 3 days.Cell proliferation was evaluated by MTT assay.Cell cycle and apoptosis were determined by flow cytometry(FCM).It is resulted that different concentrations of 1,25(OH) 2D3 could inhibited the growth of HO8910 cells and in a time and dose-dependent manner(P 0.05).1,25(OH) 2D3 could change cell cycle of HO8910 cells,increased the rates of G1,decrease therates of S and G2/M(P0.01),and so decreased cells proliferation index(PI﹡)(P 0.01).They could induced apoptosis(P0.05),then inhibit proliferation of ovarian cancer cells.It is conclused that 1,25(OH)2D3could inhibit the proliferation of human ovarian cancer cell line H08910,change the cell cycle and induced G1 phase arrest,induce apoptosis,then inhibit proliferation of HO8910 cells.
