With a special anatomical location, intestinal epithelial layer is an important barrier against antigen invasion. Its immunomodulatory role in mucosal immunity attracted considerable attention. When epithelial cells contact with pathogenic microorganisms or they are injured, the epithelial cells could interact with immunocyte in mucosal lamina propria,to recruit inflammatory cells by producing chemokines, cytokines,or by the ways of directly activated and transdifferentiation,to regulate the inflammatory or immune response in microenvironment. In this sense, intestinal epithelial layer is not only a physical barrier, but also a hinge of signal transduction in exterior and interior environment, also plays an important role in regulating intestinal mucosal inherent immunity and keeping the balance of injury and repair. As the main clinical manifestations of bowel disease, especially in inflammatory bowel disease, mucosal injury and ulcer associate closely with the changes of intestinal barrier function. So,based on the biological function and immune regulation of intestinal epithelial cells, further discussion could offer new ideas and strategies on inflammatory bowel disease-related research and clinical diagnosis.
