消化道肿瘤外周血循环肿瘤细胞与上皮间叶转变的相关性研究

摘要 目的研究循环肿瘤细胞(circulating tumor cells,CTCs)与上皮间叶转变(epithelial mesenchy-mal transition,EMT)及干细胞的关系,探寻肿瘤发生与转移的机制。方法利用葡聚糖硫酸钠(dextran sul-fate sodium,DSS)结肠癌模型C57小鼠,采集全血,用免疫磁珠负性富集方法筛选循环肿瘤细胞,并且鉴定CTCs。应用免疫荧光细胞化学染色从细胞水平鉴定Snail、N-cadherin的表达及干细胞的标志。结果检测外周血中的上皮细胞的标志pan-CK+和CD45-,确定CTCs的存在。EMT的标记Snail和N-cadherin在细胞水平的表达升高,并且鉴定到肿瘤干细胞标志的细胞。结论外周血中的循环肿瘤细胞有些具有干细胞特性,并且可能通过EMT过程扩散到循环系统中,表明CTCs与肿瘤的发展和转移有密切的关系。 Objective To study the relationship between epithelial mesenchymal transition(EMT) and circulating tumor cells(CTCs),and to explore the mechanism of tumorigenesis and metastasis.Methods We collected blood from the dextran sulfate sodium(DSS) model of C57 mice colon cancer,using immunomagnetic negative selection method to enrich circulating tumor cells,and to identify the CTCs.Immunofluorescence staining detection at the genes and the cellular level.The expression level of Snail,N-cadherin and symbol of stem cells might be associated with CTCs which have been suggested to be the active source of metastatic spread in primary.Results The epithelial cells in the peripheral blood signs pan-CK+ and CD45-determine the presence of CTCs.EMT markers such as Snail and N-cadherin at the cellular expression level are increasing.Cancer stem cell markers are identified in the cells.Conclusion The circulating tumor cells in peripheral blood with some characteristics may spread through the EMT process in the circulating system,suggesting that CTCs in tumor development and metastasis are closely related,and are critical for our molecular understanding of cell migration and morphogenesis.
