摘要
目的探讨子宫内膜癌(EC)组织中CD44拼构变异体6(CD44v6)、基质金属蛋白酶-2(MMP-2)及其抑制剂-2(TIMP-2)的表达,分析其与EC浸润、转移的关系。方法用免疫组化技术对73例EC组织和27例正常增生期子宫内膜组织中MMP-2、TIMP-2及CD44v6的表达情况检测分析。结果 MMP-2、TIMP-2及CD44v6在EC中的阳性表达均显著高于对照组(P<0.05)。子宫内膜癌中:MMP-2和TIMP-2的表达仅与肌层浸润有关(P<0.05);CD44v6与肌层浸润深度有关(P<0.05),CD44v6的阳性表达率随淋巴结转移和病理分级增高有下降的趋势,但无统计学意义(P>0.05)。MMP-2和TIMP-2及MMP-2和CD44v6在EC组织中的表达呈正相关(P<0.05)。结论 MMP-2、TIMP-2及CD44v6高表达的EC患者更易发生浸润、转移。联合检测MMP-2、TIMP-2及CD44v6的表达,可能对预测EC的转移和预后评估有重要意义。
Objective To investigate the expressions of CD44 splice variant 6(CD44v6) and matrix metalloproteinase-2(MMP-2) and its tissue inhibitor-2(TIMP-2)in endometrial carcinomas and their significance in the invasion and metastasis of the carcinoma.Methods Immunohistochemical method was used to measure the expression of MMP-2and TIMP-2 andCD44v6 in 73 case of human endometrial carcinoma and 27 normal hyperplasia endometrium tissue.Results The positive rates of MMP-2 and TIMP-2 and CD44v6 expression in endometrial carcinoma were higher than that in normal hyperplasia endometrium tissue(P0.01).The high MMP-2 and TIMP-2 expressions only related to myometrial invasion(P0.05).The higher expression of CD44v6 in carcinoma tissues was associated with greater extent of myometrial invasion(P0.05).The expression of CD44v6 in endometrial carcinoma showed decrease trend with differentiation and lymph nodes metastasis,but did not reach the statistic value(P0.05).in the expressions of MMP-2 was positively correlated to that of CD44v6 and TIMP-2 in endometrial carcinoma tissues(P0.05).Conclusion The high expressions of MMP-2 and TIMP-2 and CD44v6 in endometrial carcinoma tissues suggest their roles in the development,invasion and metastasis of endometrial carcinoma.The detection of MMP-2 and TIMP-2 and CD44v6 in combine may be a biological index in judging the malignancy,predicting the metastasis and evaluating the prognosis of endometrial carcinoma.
