雷公藤内酯醇对大鼠大脑皮质内注射Aβ后IL-1α和IL-1β表达的影响 被引量:2

摘要 目的探讨雷公藤内酯醇(TP)对大鼠大脑皮质内注射β-淀粉样蛋白(Aβ)后白细胞介素-1α(IL-1α)和白细胞介素-1β(IL-1β)表达的影响。方法 30只SD雄性大鼠等分成对照组、Aβ组、用药组。应用免疫组织化学、RT-PCR方法检测各组大鼠大脑皮质IL-1α和IL-1β的表达。结果免疫组织化学染色显示,用药组IL-1α和IL-1β阳性细胞数较Aβ组明显减少(P<0.05)、平均光密度较Aβ组明显减弱(P<0.01)。RT-PCR显示,用药组IL-1α和IL-1βmRNA水平较Aβ组明显降低(P<0.05,P<0.01)。结论 TP对Aβ所致的大鼠大脑皮质IL-1α和IL-1β的过度表达有抑制作用。 Objective To explore the effects of triptolide(TP) on the expression of interleukin-1α(IL-1α) and interleukin-1β(IL-1β) after microinjection of beta-amyloid protein(Aβ) into the cerebral cortex in rats.Methods Thirty male SD rats were equally divided into control,Aβ and TP-treated groups.The variations of IL-1α and IL-1β expressions of the cerebral cortex in these groups were observed by immunohistochemical staining and RT-PCR.Results The cell numbers of IL-1α and IL-1β positive staining in TP-treated group were significantly decreased than those in Aβ group(P0.05),and the average optical densities of IL-1α and IL-1β positive staining in TP-treated group were also significantly lower(P0.01).The expression levels of IL-1α and IL-1β mRNA in TP-treated group were significantly lower than those in Aβ group(P0.05,P0.01).Conclusions TP can inhibit the overexpressions of IL-1α and IL-1β in the cerebral cortex induced by Aβ.
