大黄素致鼻咽癌细胞CNE-1胀亡和线粒体膜通透性的研究 被引量:1

摘要 目的:探讨大黄素对人鼻咽癌细胞增殖抑制、胀亡形态及线粒体膜通透性转换孔(PTP)。方法:将人鼻咽癌细胞株CNE-1分为不同药物处理组,并设立阴性对照组。采用MTT法观察大黄素对CNE细胞体外生长抑制作用,采用HE染色、透射电镜观察CNE细胞在药物作用后胀亡形态。通过紫外分光光度法观察大黄素对CNE细胞线粒体通透性影响。结果:大黄素呈浓度依赖性抑制CNE细胞体外生长(P<0.01)其IC50为206.78μg/ml。HE染色结果可见大黄素组细胞体积开始变大,细胞胞浆空泡化,胞浆内出现致密颗粒,细胞核开始变形细胞质空泡明显,电镜结果可以观察到线粒体肿胀,脊脱出,线粒体出现空泡化,表现为典型细胞胀亡。线粒体通透性转换孔开放实验发现线粒体开放程度与大黄素浓度有依赖性。结论:大黄素可以使鼻咽癌细胞走向胀亡,其途径可能与线粒体有关。 Objective To study the inhibition effect of oncosis and mitochondrial membranes permeability(PTP ) of human nasopharyngeal carcinoma cell -CNE-1 induced by Emodin.Methods CNE-1 was divided to several test groups treated with different concentrations Emodin and a control group didn't treated with Emodin.MTT method,HE and transmission electron microscope method, UV method was used to observe the inhibition effect of oncosis and mitochondria membranes permeability of CNE-1 treated by Emodin, respectively.Results the MTT method result showed that the inhibition effect of Emodin to CNE-1 was correlated its concentrations (P0.05 ),and its IC50 was 206.78μg/ml.the HE method result showed that CNE-1 became expand,cavity and dense granule advented in hyalomitome,and cell nucleus became deformities.the electron microscope method result showed that mitochondrial became expand and vacuolization,showed a typical phenomenon of oncosis.The test mitochondrial membranes permeability showed that the open degree of mitochondrial was correlated with Emodin's concentrations.Conlusions Emodin can leaded CNE-1 to oncosis,this effect may relate to the mitochondrial.
