摘要
目的:探讨Meis1在围着床期小鼠子宫内膜中的作用。方法:采用免疫组织化学定位检测Meis1在小鼠子宫内膜中的表达;采用Real-time RT-PCR和免疫印迹方法,分别在mRNA和蛋白水平定量检测Meis1在围着床期小鼠子宫内膜中的表达水平。结果:免疫组织化学结果显示,Meis1蛋白主要表达于小鼠子宫内膜腺上皮细胞的细胞膜和胞质以及基质细胞和蜕膜细胞的细胞核中,在围着床期小鼠子宫内膜中的表达随妊娠天数的增加而增加,着床后在基质和蜕膜中的表达增强;在孕第4日显著增加,孕第5日达高峰,孕第6日表达下降。Real-time RT-PCR和免疫印迹结果显示,Meis1的表达随妊娠天数的增加逐渐增强,孕第4日明显增强,孕第5日达高峰,孕第6日开始下降。结论:Meis1在围着床期小鼠子宫内膜中呈规律表达,与小鼠着床窗吻合,提示Meis1是调控围着床期子宫内膜容受性的重要因子。
Objective: To detect the expression of Meis1 in endometrium of mice in periimplantation period.Methods: Immunohistochemistry was used to detect the location and semi-quantitativity of Meis1 in endometrium of mice in periimplantation period;Respectively on the level of Meis1 mRNA and protein,real time RT-PCR and western blotting were used to detect quantitativity of in endometrium of mice in periimplantation period.Results: Immunohistochemistry showed that Meis1 expressed in the cytoplasm and membrane of edometrial glandual epithelium cells and the nucleu of endometriumal stromal and decidual cells;Real time RT-PCR and Western blotting showed that the expression of meis1 mRNA and protein was weakly expressed on pregnant day 1 and day 2,significantly expressed on day 4,achieved to the peak on day 5,it showed decent trend on day 6.Conculation: Meis1 is expressed disciplinarily in endometrium of mice during periimplantation period,which is corresponding to mice implantation window.It is implied that Meis1 is an important factor in regulat-ing endometrium receptivity during periimplantation period.
