摘要
目的观察肿瘤内置入5-FU缓释化疗粒子治疗肝癌的效果。方法 12只Balb/C小鼠随机分为对照组、5-FU缓释化疗粒子治疗组,每组6只,在每组小鼠右侧腋窝皮下接种2×106个H22肝癌细胞,10d后均成瘤,肿瘤直径大小约1cm,对照组不进行干预,5-FU缓释化疗粒子治疗组在小鼠肿瘤中心部位植入规格为2mg的缓释化疗粒子1粒,粒子植入21d后处死小鼠,测定肿瘤的长径和短径,计算肿瘤体积,计算治疗组的肿瘤体积抑制率,对肿瘤组织进行常规病理切片观察组织学变化。结论对照组5-FU缓释化疗粒子治疗组和联合治疗组小鼠在粒子植入21d后肿瘤体积分别为(14425.80±2656.91)mm3、(4997.29±538.71)mm3。与对照组比较,5-FU缓释化疗粒子治疗组体积抑瘤率为65.36%,病理学结果显示:对照组肿瘤细胞生长良好,治疗组肿瘤组织均有不同程度的细胞坏死。结论 5-FU缓释化疗粒子植入治疗肝癌的效果明显。
Objective To evaluate the treatment effect of 5-FU chemotherapy seed implantation on H22 xenograft in mice.Methods 2×106/0.2 ml H22 cell were transplanted subcutaneously in the right axillary fossa of 12 Balb/C mice,and 10 days later mice with a tumor of 10 mm in mean diameter were produced and devided into 2 group: control group and chemotherapy seed group. Each group included 6 mice. One chemotherapy seed(2 mg) was implanted into the xenograft in chemotherapy seed group, the tumor in control group was not treated. The tumor’s long diameter and short diameter in each group were measured 21 days later and the tumor’s volume were calculate according to formla V=0.52ab2(a was long diameter, b was short diameter),the tumor volume inhibiting rate in treatment group was calculated. Routine pathological slides of tumor tissue were observed under light microscope. Results 21 days later the average xenograft volume in control group was(14425.80±2656.91)mm3, while in chemotherapy seed implantation group was 4997.29±538.71 mm3. The tumor volume inhibition rate in chemotherapy seed implantation group was.65.36%.Conclusion 5-FU chemotherapy seed implantation can inhibit the growth of tumor and can be used in liver cancer.
