摘要
目的:评价重组人胰岛素样生长因子1(recombinant Insulin-like growth factor-1,rhIGF-1)对新生儿缺氧缺血性脑病(hypoxic ischemic encephalopathy,HIE)模型大鼠的认知功能、海马细胞形态学变化及Akt1 mRNA表达量的影响,为临床上治疗HIE提供理论依据。方法:选取7日龄SD大鼠15只,随机分为模型组、治疗组和对照组,运用缺血灌注和缺氧处理模型组和治疗组大鼠,建立HIE模型。治疗组给予rhIGF-1鼻吸处理2次(间隔1 h)。15 d后,用Morris水迷宫观察大鼠逃避潜伏时间(s),观察海马区CA1区细胞数量,运用RT-PCR方法检测Akt1表达量变化。结果:与模型组大鼠相比,治疗组大鼠学习记忆能力明显提高,海马CA1区细胞数量明显增多,海马区Akt1 mRNA表达明显增多(F检验,P<0.05),模型组上述指标均低于对照组(F检验,P<0.05)。结论:rhIGF-1可以明显改善HIE大鼠认知功能,增加海马区Akt1 mRNA表达,有明显的脑保护作用。
Objective:To evaluate the effect of recombinant human insulin-like growth factor-1(rhIGF-1) on cognitive function in rats with hypoxic-ischemic encephalopathy(HIE) and to provide evidences for treatment.Methods:15 SD rats of 7-day-old were randomly divided into model group,treatment group and control group.Established HIE model,model group and treatment group rats were treated with monolateral carotid ligation and hypoxia;at the same time treatment group received rhIGF-1 by intranasal administration(2 times,interval 1 hour).After 15 days,the cognitive function of rats were measured with Morris water maze test,and number of cells,Akt1 mRNA expression in hippocampal CA1 sector were tested.Results:Compared with model group,the cognitive function of escape latency in the maze test were increased significantly(P0.05);and number of cells,Akt1 mRNA expression in hippocampal CA1 sector were also increased in treatment group(P0.05).Conclusion:RhIGF-1 can significantly improve cognitive function in rats with HIE.,enhances the Akt1 mRNA expression in hippocampal CA1 sector,and have protective effects on brain.
