摘要
目的:观察为期16天的预热适应对大鼠一次性离心运动后骨骼肌损伤的保护作用及对自由基代谢的影响。方法:5周龄SPF级雄性SD大鼠80只,随机分为非预热适应组和预热适应组。并根据取材点不同将非预热适应组和预热适应组分别分为非预热适应安静对照组、单纯运动后即刻、24 h、48 h组、6 d组;预热适应安静对照组和预热适应运动后即刻、24 h、48h、6 d组。为期16天的预热适应结束后,室温恢复24 h,除非预热适应安静对照组和预热适应安静对照组外,运动组大鼠均进行一次性力竭离心运动。运动结束后,分别在运动后即刻、24h、48 h和6 d取腓肠肌,进行光镜观察和SOD、MDA含量测试。结果:1)光镜下观察到,预热适应大鼠运动后骨骼肌损伤较单纯运动大鼠相应的时间点明显减轻。2)预热适应大鼠运动后SOD活性较单纯运动大鼠同一时间点的值均高,其中预热适应运动后24 h显著高于单纯运动后24 h(P<0.01);而MDA含量较单纯运动大鼠同一时间点的值均低,其中预热适应运动后24 h、48 h与单纯运动后同一时间点比较,差异有显著性(P<0.01和P<0.05)。结论:为期16天的预热适应对离心运动所致骨骼肌损伤有明显的保护作用,并能纠正其自由基代谢失调。
Objective:to investgate whether heat pretreat adaptation of 16 days had protective in rats against skeletal muscle injury induced by an acute eccentric exercise and the effect of free radical metabolism.Methods:80 SPF-class male SD rats of 5-week old were randomly divided into non-preheat adaptated groups and preheat adaptated groups.These groups were normal sedentary control group(C),immediately,24 hours,48 hours,6 days after simple exercise(E0,E1,E2 and E6),preheat adaptation quiet group(HC),immediately,24 hours,48 hours,6 days after heat pretreat exercise(HE0,HE1,HE2,HE6).Rats were allowed to recovery from the last whole-body hyperthermia for 24 hours.All the exercise groups performed an exhaustive exercises on a treadmill.The rats were killed on 0,1,2 and 6 days after exercise and isolated gastrocnemius,which were sampled to observed by light microscopy and analyze SOD and MDA.Results:1) Skeletal injury degree of Groups HE were largely lower than that of Groups E.2) SOD isolated gastrocnemius,activity in Groups HE were higher than that of Groups E each time.Its activity of Groups HE1 was significantly higher than that of Group E(P0.01);MDA content in Groups HE were lower than that of Group E each time,Its content of Group HE1,Group HE2 markedly lower than that of Group E1 and Group E2(P0.01and P0.05,respectively).Conclusion:The 16-day pre-heat adaptation can protect against skeletal muscle injury induced by eccentric exercise,and correct free radical metabolism disorder in skeletal muscles.
