猪传染性胃肠炎病毒陕西株的分离鉴定 被引量:12

Isolation and identification of porcine transmissible gastroenteritis virus(TGE) shaanxi strain and sequence analysis of its N gene

摘要 从陕西省某猪场临床上表现为腹泻症状的病死仔猪肠道内容物及粪便材料中分离获得1株病毒,采用PK-15细胞培养、理化试验及中和试验鉴定后,证实该分离株为猪传染性胃肠炎病毒(TGEV)。克隆分离株N基因,并将该基因核苷酸序列和推导的氨基酸序列与其他不同来源的TGEV毒株进行比较分析。结果表明,分离株N基因与其他毒株核苷酸的同源性为94.3%~98.3%,氨基酸同源性为95.6%~99.4%。系统进化树结果表明,分离株与TGEV HN2002株(河南株)亲缘关系最近。 A TGE virus strain was successfully isolated from intestinal tract contents and excrement of died piglets with clinical diarrhea symptom from a pig farm in Shaanxi Province,cultured in PK-15 cell culture, and examined by physicochemical test and neutralization test (NT). A DNA fragment about 1149 bp of TGEV isolated from Shaanxi which contained the ORF of N gene was cloned with RT-PCR. The N gene of Shaanxi strain was amplified by PCR which shared 94.3%-98.3% homology of nucleotide sequences and 95.6%-99.4% homology of amino acids with those of 10 TGEV strains. The results of the phylogenetic tree analysis demonstrated that there was a close genetic relation ship between Shaanxi strain and HN2002 strain, and they are in the same branch. The result indicated that the differences between different strains were not obvious.
