基于secY基因对山东临沂枣疯病植原体的分子鉴定 被引量:5

Molecular Identification of Phytoplasma Associated with Jujube Witches' Broom in Shandong Province Based on secY Gene

摘要 利用FD9f/r引物对采自山东临沂的枣疯病样品进行secY基因的PCR扩增,并进行了序列测定,结果表明,该特异片段大小为1 421 bp。对获得的序列与NCBI数据库中相关植原体序列进行同源性分析、构建系统进化树和模拟RFLP分析,结果显示山东临沂枣疯病植原体属于secYⅤ-C亚组,与secYⅤ-B亚组的樱桃致死黄化株系(CLY5)和secYⅤ-N亚组的桃树致死黄化印度分离株系(PY-IN)的亲缘关系最近,在亚组水平上进一步明确了山东临沂枣疯病植原体的分类地位。 The secY gene of jujube witches' broom phytoplasma (JWB) collected from Linyin in Shandong Province was amplified using FD9f/r primers and sequenced. The obtained fragment was 1 421 bp. The sequence of jujube witches' broom phytoplasma was compared with the related sequences of phytoplasma in NCBI. The results of phylogenetic analysis and virtual RFLP showed that jujube witches' broom phytoplasma belonged to subgroup secYV - C, having the closest relationship with CLY5 (secYV - B) and PY - In ( secY V -N).
