摘要 目的:改进传统制作血液涂片技术,使其统一标准化,有效控制血液检验的质量。方法:制作10°~30°不同固定角度推片,用一定量的EDTA抗凝血制作血液涂片,观察血膜长度及染色后显微镜下细胞分布均匀度,并进行测量比较,找出符合制备标准角度的推片,制备标准血液涂片,与传统的制片法制作的血液涂片进行比对分析。结果:17°或18°固定角度推片,用血量5uL制作血液涂片,血膜长度X和S为4.081,3.987;0.015,0.0103;细胞分布均匀区X,S为3.033,2.971;0.015,0.0104。17°固定角度推片和传统的制备方法比较P<0.001,有显著性差异。结论:17°或18°固定角度推片,用血量5uL制作的血液涂片符合标准血液涂片,在细胞形态检查中能有效进行质量控制,加大了对疾病诊断的准确率。 Objective:To improve the traditional method of making blood smear slides to standardize the new method to effectively control the quality of blood tests.Methods:Using push-slides at an fixed angle of different degrees from 10° to 30° to make blood smear slides with a certain amount of EDTA anticoagulated blood.Under a microscope,the length of the blood membranes and the evenness of the distribution of cells were observed after staining the smear slides.Then measurement and comparison were performed to find out those smear slides which conform to established standards and identify the standard angles of push-slides.The standard blood smear slides were made using push-slides at the standard angles.The standard blood smear slides were compared with those made by the traditional method.Results:The blood smear slides were made with 5uL of blood using push-slides at an fixed angle of 17°/18° degree.The X and S values of membrane lengths for the slides were 4.081/3.987 and 0.015/0.0103,respectively.The X and S values for the areas of evenly distributed cells for the slides were 3.033/2.971 and 0.015/0.0104,respectively.The P value for the comparison between the slides by the traditional method and those by using push-slides at a fixed angle of 17° was less than 0.001,which means there is significant difference between these 2 kinds of slides.Conclusion:Blood smear slides made by using push-slides at a fixed angle of 17° or 18° with 5uL of blood satisfies the requirements for standard blood smear slides,and are therefore helpful in controlling the quality of morphological examination of blood cells,thus increasing the accuracy of diagnosis.
