不同剂量钴-60辐照对制痂酊中儿茶素和表儿茶素含量的影响 被引量:9

Effects of Different Dose (60)~Co Irradiation on the Contents of Catechin and Epicatechin in Zhijia Tincture

摘要 目的:建立HPLC法测定制痂酊中儿茶素和表儿茶素含量的方法,同时考察不同剂量^(60)Co辐照对其含量影响。方法:色谱柱:Kromasil C_(18)(250 mm×4.6 mm,5μm),流动相:2.2%冰醋酸-乙腈(86:14),流速:0.8 ml·min^(-1),检测波长:280nm,柱温:35℃。结果:儿茶素和表儿茶素分别在1.025~12.813μg,0.196~2.450μg范围内线性良好,加样回收率分别为98.0%,99.8%(n=5);^(60)Co辐照剂量为3 kGy时,儿茶素和表儿茶素含量分别减少2.77%,3.43%,随着辐照剂量的增加,两者含量有逐渐下降的趋势。结论:对单批次样本测定表明:^(60)Co辐照对制痂酊中儿茶素和表儿茶素的含量有一定影响,但在低剂量(≤3 kGy)时影响不显著。 Objective : To establish an HPLC method for the simultaneous determination of eateehin and epieateehin in Zhijia tincture and study the effects of different dose 6o Co irradiation on the contents. Method: Kromasil C18 column (250 mm× 4. 6 mm,5 μm) was used. The mobile phase consisted of 2. 2% aeetum-aeetonitrile (86: 14) with the flow rate of 0. 8 ml·min^-1. The detection wavelength was set at 280 nm and column temperature was 35 ℃. Result: Good linearity was obtained over the range of 1. 025-12. 813 μg for catechin and 0. 196-2. 450 μg for epicatechin with the average recovery of 98.0% and 99. 8% , respectively (n = 5 ). The content of eateehin and epieateehin in Zhijia tincture (irradiated by 3 kGy of ^60Co) was decreased 2. 77% and 3.43% ,respectively,and the contents had declining trend with the increase of irradiation dose. Conclusion:^ 60Co irradiation has influence on the contents of eatechin and epieateehin in Zhijia tincture, however the influence is not significant when the irradiation dose is less than 3 kGy.
