摘要
本文分离纯化了正常大白鼠肝脏和癌前病变大白鼠肝脏组织线粒体谷胱甘肽S-转移酶(GST),对其部分性质进行了探讨。经O-(二乙胺基乙基)-纤维素、羟基磷灰石,羧甲基-琼脂糖6B(CM-Sapharose 6B)柱层析以及等电聚焦电泳法,从正常肝组织和癌前病变肝组织中,分离纯化了两种不同性质的GST,暂命名为线粒体GST_1和线粒体GST_2。通过十二烷基磺酸钠(SDS)-聚丙烯酰胺凝胶电泳法和等电聚焦电泳法测定的线粒体GST_1的分子量约为23000,等电点为6.2,线粒体CST_2的分子量约为45000,等电点为9.1。在正常组织和癌前病变组织中CST同工酶无质上的差异。但是,线粒体GST_1在癌前病变组织中的活性明显著高于正常对照组(P<0.01)。
Mitochondrial GST of normal liver and hepatocyte nodules of rats were purified and some properties were identified. Two isoenzymes of mitochondrial GST were purified by sequential chromatography on DEAE cellulose, hydroxylapatite and CM-Sepharose 6B finally by polyacrylamide gel isoelectric focusing from hepatocyte nodules and normal liver tissue, which are temporarily named mitochondrial GST_1 and mitochondrial GST_2. The molecular weight of mitochondrial GST_1 subunit is approximately 23000, and that of mitochondrial GST_2 subunit, 45000, which are all SDS-polyacrylamide disc gel electrophoresis and isoelectric point of mitochondrial GST_1 is 6.2 and that of mitochondrial GST-2, 9.1, which are all determined by the polyacrylamide gel isoelectric focusing. The property of mitochondrial GST_1 is not different from that of GST_2, but activities of mitochondrial GST_1 in nodules increased significantly higher than that of normal liver tissue.
