眼针对大鼠脑缺血再灌注损伤72h后脑皮质BDNF表达的影响 被引量:4

The Effect of Eye Acupuncture on Brain-deprived Neurotrophic Factor Expression in Rat cerebral cortex with Ischemia-reperfusion injury for 72 hours

摘要 目的:观察眼针对脑缺血再灌注损伤大鼠脑皮质脑源性神经营养因子(BDNF)表达的影响。方法:线栓法复制脑缺血再灌注损伤大鼠模型,随机分为正常组、假手术组、模型组、眼针组。于再灌注72h后进行大鼠神经功能评分,采用RQ-PCR、免疫组化法、western blot法检测缺血脑皮质BDNFmRNA及蛋白的表达。结果:与模型组比较,眼针组大鼠神经功能评分下降,脑皮质BDNF mRNA的表达和蛋白的表达上升(P<0·01)。结论:眼针能提高脑缺血再灌注损伤大鼠脑皮质BDNF表达水平。 Objective:To investigate the effects of eye acupuncture on brain-deprived neurotrophic factor(BDNF) expression and study the mechnism of ischemia-reperfusion injury improved.Methods:Rats established by suture method were randomly divided into the control group,the sham operation group,the model group and the eye-acupuncture group.After reperfusion 72h,the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks;the expression of ischemic cerebral cortex BDNF mRNA was measured by RT-PCR method;the expression of ischemic cerebral cortex BDNF protein was detected by immunohisto-chemistry and Western blot technology.Results:After reperfusion 72h,compared with the model group,the neurologic impairment marks of the eye-acupuncture group decreased obviously(P0.05);The expressions of BDNF mRNA and protein in rat cerebral cortex after the eye acupuncture therapy were obviously decreased(P0.01).Conclusion:The eye acupuncture therapy can increase the expression of BDNF in rat cerebral cortex with ischemia-reperfusion injury,and protect brain by improving the repair of neurons.
