摘要
本文对鼠伤寒沙门氏菌致突变试验在国内实验条件下方法标准化进行了研究。国产试剂配制的营养肉汤可满足试验菌株的营养需求,烧瓶振荡培养10小时即可达1~2×10~9存活菌/ml,肉汤培养物稀释后测定OD650可监测细菌密度。国产五氯联苯和Aroclor 1254诱导的Sprague-Dawley及Wistar大鼠肝S9活化苯并(a)芘及2-乙酰氨基芴的能力相似(P>0.05)。本文还提出以点试验估计受试物毒性及以参比物减小或消除多次实验间的误差。
The standardized procedure of Salmonella mutagenicity test was studies. The nutrient broth prepared from the home-made reagents was used for the growth of tester strains. Tester strain cultures in flask shaked at 100-120 rpm were grown to a density of 1 ~ 2 x 109 cells per ml in 10 hrs.The tester bacteria density may be monitored by turbidity measurements at 650 nm. Comparision studies for detecting differences between hepatic S9 from Sprague-Dawley and Wistar rats were carried out with pretreatment of Aroclor 1254 and home-made pentachlorobi-phenyl. These hepatic S9 showed the similar metabolic activation capacities on benzo(a) pyrene and 2-amenylaminofluorene to directly acting mutagens ( P>0.05 ) . Aroclor 1254 and pentachlorobiphenyl increased the cyrochrome P450 content,aryl hydrocarbon hydroxylase and N-demethylase activities of these S9 preparation, but aryl hydrocarbon hydroxylase activity of liver S9 from Wistar rat induced by Aroclor 1254 was higher than that from Sprague-Dawley rat. Standardized procedures for estimating toxicity of test compound with spot test and for reducing the daily variations of test results with a referance mutagen were proposed.
