三氯蔗糖-6-乙酯的制备及结构表征 被引量:3

摘要 以蔗糖为原料,原乙酸三乙酯为酯化剂,经C-6乙酰化,生成蔗糖-6-乙酯,再用氯化亚砜和DMF制备的Vilsmeier试剂与蔗糖-6-乙酯进行氯化反应,得到三氯蔗糖-6-乙酯。考察了反应物的投料比对酯化反应的影响;反应温度、反应物的投料比和反应时间等因素对氯化反应的影响。结果表明:最佳反应条件为酯化反应温度为0℃,反应时间4 h,n(蔗糖)∶n(原乙酸三乙酯)∶n(对甲苯磺酸)=1∶2.5∶3.0×10-2,收率为70.2%。氯化反应温度110℃,反应时间2 h,n(蔗糖-6-乙酯)∶n(Vilsmeier)=1∶18.5,收率为51.9%。通过HPLC-MS、IR、1H NMR和13C NMR对产物的结构进行了表征。 Sucralose-6-ester was prepared by the reaction of C-6 acylation and chlorination with Vilsmeier reagent generated from thionyl chloride and DMF,using sucrose and triethyl orthoacetate as the raw materials.The effect of the molar ratio of raw material on acetylation was investigated.The effects of reaction temperature,the molar ratio of raw material and reaction time on chlorination were also investigated.The optimum conditions of acetylation were as follows:the reaction temperature,0 ℃;the reaction time,4 h and n(sucrose)∶n(triethyl orthoacetate)∶n(p-toluene sulphonic acid)=1∶2.5∶3.0×10-2.The yield was 70.2%.The optimum conditions of chlorination were as follows: the reaction temperature,110 ℃;the reaction time,2 h and n(sucrose-6-ester)∶n(Vilsmeier)=1∶18.5.The yield of sucralose-6-ester was 51.9%.Sucralose-6-ester was characterized by HPLC-MS,IR,1H NMR and 13C NMR.
