摘要
目的 PCR扩增ICR小鼠脂肪组织的resistin基因,为进一步在ICR小鼠中开展resistin的相关研究提供必要的基础。方法在脂肪组织中抽提总RNA,用RT-PCR进行resistin基因的体外扩增,经电泳检测和测序后用DNAMAN软件进行碱基序列分析。结果抽提的总RNA完整性较好,RT-PCR产物电泳检测获得了363 bp的目的条带,双向测序的碱基峰值图均一,所扩增片段的序列与GenBank报道的小鼠resistin序列有93.8%的同源性,说明获得的基因片段是ICR小鼠的resistin基因。结论在体外成功扩增了ICR小鼠脂肪组织的resistin基因,可为在ICR小鼠中开展resistin基因生物功能等的研究奠定基础。
Objective To amplify the resistin gene from adipose tissue of ICR mice for further research of its functions.Methods Total RNA was extracted from the adipose tissue for amplification of resistin gene by RT-PCR.The sequence of amplified nucleotide acids was analyzed by DNAMAN software after gel electrophoresis and sequencing.Results The total RNA had good integrity.The expected 363 bp fragment was obtained by RT-PCR.The base peak was uniform in both directions of sequencing.The amplified fragment showed a 93.8% homology with the reference of mouse resistin gene in GenBank,indicating that the resistin gene of ICR mice was obtained.Conclusion The resestin gene of ICR mice has been successfully amplified from adipod tissue,which laid a foundation for further studies on biological functions of resistin gene in ICR mice.
