摘要
目的:探讨基因沉默MTA1对宫颈癌CaSki细胞株生物学效应的影响。方法:构建针对MTA1基因的siRNA,体外转染人宫颈癌CaSki细胞株,通过RT-PCR检测其对CaSki细胞MTA1 mRNA表达的影响,并检测转染前后细胞增殖、生长及细胞周期改变情况。结果:瞬时转染MTA1-siRNA入CaSki细胞,可见细胞中MTA1 mRNA表达降低,转染后细胞生长减慢,克隆形成及划痕愈合能力下降,但对细胞周期的改变不明显。随着干扰浓度的增加,时间的延长,siRNA对MTA1 mRNA表达的抑制作用逐渐增强。结论:基因沉默MTA1能使宫颈癌细胞生长减慢,细胞迁移能力下降,一定程度上逆转了宫颈癌细胞的恶性表型。
Objective:To investigate the effect of RNA interference against MTA1 on the biological behaviors of cervical cancers.Methods:We synthesized siRNA targeting MTA1 gene,then transfected it into human cervical cancer cell line CaSki.RT-PCR was used to analyse MTA1 mRNA expression levels while cell proliferation,growth,and cell cycles were also examined simultaneously.Results:The study showed that RNA interference against MTA1 played its role to decrease MTA1 expression in CaSki cells.The biological behaviors of cell growth,colony formation and wound healing ability all decreased,but it had little effect on the cell cycles with the increase of RNA interference concentration and the extension of function duration,the expression of MTA1 mRNA decreased gradually.Conclusion:Gene silencing MTA1 can decrease cancer cell growth and migration.The general outcome of RNAi against MTA1 is to reverse the malignant phenotype.
